No — dogs are not allowed
No dogs anywhere on the Virginia end of Assateague — including inside your vehicle. This is one of the strictest pet bans on the east coast and both managing agencies state it plainly: the National Park Service says "Pets are prohibited in the entire Virginia portion of Assateague Island, even in your car", and the U.S. Fish & Wildlife Service, which manages nearly all of that land as Chincoteague National Wildlife Refuge, lists "Pets, even in your vehicle" as prohibited. You cannot leave a dog in the car park while you walk the beach, and there is no seasonal exception. ADA service animals are permitted, on a leash of 6 feet or less. If you want to see Assateague with your dog, go to the MARYLAND district instead, where pets are allowed in specific areas on a 6-foot leash (not on nature trails, lifeguarded beaches, or backcountry campsites) — but note the two ends of the island are not linked by road, so it is a separate drive of roughly three hours, not a short hop.
Where dogs ARE allowed
Nowhere on the Virginia side — not the beach, not the trails, not the car park, not inside your locked car. Only ADA service animals are exempt. The nuance that matters for trip planning is the OTHER end of the same island: "Pets are permitted only in specific areas of the Maryland portion of the National Seashore", on a leash "no longer than 6 feet at all times", and still barred from backcountry campsites, nature trails, lifeguard-protected beaches, and the stretch north of the State Park to the Ocean City inlet. Maryland and Virginia are not connected by road within the island — reaching the Maryland district is a roughly 3-hour drive around.
